Peptide immunization

Peptide immunization uses synthetic, screened epitope peptides to trigger targeted adaptive immunity through MHC presentation. Ideal for vaccines, tumor immunotherapy and antibody development, it outperforms traditional whole-protein, bacterial and cell-based immunization methods with unique strengths.

Peptide immunization elicits specific immune responses based on precisely screened antigenic epitope peptides, with five core advantages as follows:

  • Superior targeting precision. It can distinguish protein modifications, isoforms and mutation sites, and simultaneously activate dual immunity consisting of humoral response and cytotoxic T cell response.
  • Low safety risks. Peptides are purely chemically synthesized with no risk of infection, and free from severe allergies or off-target inflammation induced by impure foreign proteins.
  • Flexible design and modification. Tandem multi-epitope construction and site-specific modification enable rapid multi-target combined immunization, solving the challenges of immunizing hard-to-express proteins.
  • Simplified production and quality control. The synthesis process delivers stable performance and excellent batch consistency, with lower barriers for storage, transportation and overall costs.
  • Favorable small-molecule permeability. Peptides are readily taken up by immune cells. 1st: 0.5 mg peptide immunogen + Complete Freund's Adjuvant (CFA); Subcutaneous injection at 10 sites.

2nd: 0.25 mg peptide immunogen + Incomplete Freund's Adjuvant (IFA); Subcutaneous injection at 5 sites.

Day 21

Collect 1 mL of whole blood, separate serum, and perform ELISA titer testing.

3rd: 0.25 mg peptide immunogen + Incomplete Freund's Adjuvant (IFA); Subcutaneous injection at 5 sites.

Collect 1 mL of whole blood, separate serum, and perform ELISA titer testing.

Day 42

4th: 0.25 mg peptide immunogen + Incomplete Freund's Adjuvant (IFA); Subcutaneous injection at 5 sites.

Day 49

Collect 10 mL of whole blood, separate serum, and perform ELISA titer testing. Based on titer results:

  • Collect 50 mL of whole blood, separate serum, perform protein ELISA titer testing. 
  • From the remaining whole blood, isolate PBMCs, lyse 2.0E+07 cells with 1 mL TRIzol, and aliquot at 1 mL per tube.
