Cell Immunization

Cellular immunization is an immunization technique that uses whole-cell materials including viable intact cells, fixed cells, cell lysates and cell membrane fractions as immunogens to directly immunize laboratory animals. It stimulates the animal’s immune system to generate polyclonal or monoclonal antibodies targeting cell surface antigens, intracellular proteins and cell-specific markers.

Cell antigen immunization is a technique for generating antibodies against cellular markers by directly immunizing animals with whole cells, cell lysates or cell membrane fractions as natural complete antigens. Unlike small-molecule haptens, cells inherently contain numerous immunogenic epitopes such as native proteins, glycoproteins and lipopolysaccharides, which carry intrinsic recognition sites for T cells and B cells. No carrier protein conjugation is required, making cells natural complete antigens. Following immunization, the animal produces a diverse mixture of antibodies that target distinct cellular components, including cell membrane receptors, differentiation antigens, intracellular cytoskeletal proteins and tumor-specific antigens.


The primary advantage of this approach lies in its preservation of the native spatial conformation of antigens, facilitating the generation of functional antibodies suitable for flow cytometry and immunofluorescence assays. It also eliminates the processes of protein purification and hapten conjugation. Its main drawback is the abundant non-specific antibodies generated post-immunization, which lead to high background interference. High-specificity antibodies against low-abundance cellular targets are difficult to obtain. This method is widely applied in the screening and development of antibodies targeting cell surface markers, tumor cells and immune cells.


Common types of cellular antigens include tumor cells, immune cells (T cells, B cells, macrophages), stem cells, pathogenic microbial cells (bacteria, fungi), and genetically engineered stable cell lines.

Collect 10 mL of whole blood as reserved negative serum.

Day 1

1st: 1E8 cell immunogen + Complete Freund's Adjuvant (CFA); Subcutaneous injection at 10 sites.

Day 14

2nd: 1E8 cell immunogen + Incomplete Freund's Adjuvant (IFA); Subcutaneous injection at 5 sites.

Day 21

Collect 1 mL of whole blood, separate serum, and perform ELISA titer testing.

Day 28

3rd: 1E8 cell immunogen + Incomplete Freund's Adjuvant (IFA); Subcutaneous injection at 5 sites.

Day 35

Collect 1 mL of whole blood, separate serum, and perform ELISA titer testing.

Day 42

4th: 1E8 cell immunogen + Incomplete Freund's Adjuvant (IFA); Subcutaneous injection at 5 sites.

Day 49

Collect 10 mL of whole blood, separate serum, and perform ELISA titer testing. Based on titer results:

  • Collect 50 mL of whole blood, separate serum, perform protein ELISA titer testing. 
  • From the remaining whole blood, isolate PBMCs, lyse 2.0E+07 cells with 1 mL TRIzol, and aliquot at 1 mL per tube.
